Description Cystatins, the inhibitors of cysteine peptidases, are expressed and released by many parasitic organisms. They are participating on regulation of biological processes, e.g., the modification of the host immune response. Our study is primarily focused on characterization of molecular, biochemical and immunological properties of cystatins of four model species representing the four main groups of helmints: Fascioloides magna (Trematoda), Hymenolepis diminuta (Cestoda), Diplozoon paradoxum (Monogenea) and Trichinella spiralis (Nematoda). The excretory-secretory products were isolated, purified and fractionated and the identification of cystatins is in process. The RNA was isolated, reverse transcription performed and prepared cDNA used in PCR with specific primers designed for amplification of particular cystatine genes. Obtained amplicons will be sequenced and the selected cystatine gene variants will be cloned and prepared in recombinant form for further characterization.
