p53 tumor suppressor inhibits proliferation and induces differentiation of v-myb-transformed monoblasts

Description | The aim of this study was to investigate whether human p53 protein can suppress transforming activity of the v-Myb oncoprotein in BM2 cells. Therefore, we transfected BM2 monoblasts with cDNA coding for human p53 and evaluated phenotype of p53-expressing BM2 cells of several independent clones of stable transfectants. Increased p53 level caused growth arrest of BM2p53 cells in G2-phase of the cell cycle. At the same time, the p53 protein resumed terminal differentiation capability of BM2 cells as documented by increased adherence, phagocytic activity, production of reactive oxygen species and alpha-naphtyl acetate esterase activity. These results show that p53 tumor suppressor protein can suppress transformation the v-myb oncogene in leukemic cells. |
