Description Spirochete T. pallidum ssp. pallidum, the causative agent of syphilis, is an obligate human pathogen that cannot be cultured continuously under in vitro conditions. Genome sequencing and constructions of BAC library of treponemal DNA in E. coli open new areas for the next studies (e.g. for the study on gene expression regulation). Genes rpoN (coding for 54 factor) and fhlA (coding for putative transcriptional activator) were cloned into the arabinose-inducible expression vector pBAD/HisB. Q-PCR technique was used for the study of putative gene regulation mediated by RpoN and FhlA in the clones of the BAC library. 54-dependent promoters were identified in the T. pallidum genome using in silico approach and the predicted transcripts were mapped by PCR technique. Proteins RpoN and FhlA are positive regulators of gene expression of several treponemal genes.
