Description | Colicins are antibacterial toxins produced by strains of Escherichia coli and related bacteria that provide a selective advantage against closely related strains in the competition among bacteria. Toxic effect that is mediated by the C-terminal domain is depolarization of the cytoplasmic membrane of sensitive cells. The plasmid encoded immunity protein ensures that producers are protected against the effect of their own colicin. The recognition of soluble colicin by the membrane-bounded immunity protein is a suitable model for studying protein-protein interactions. The pair of colicins U and Y had 87% identity in their pore-forming domains but the corresponding producers were not cross-immune. The amino acids of colicin U (Y) were mutated into those appearing in the sequence of colicin Y (U) and changes in the interactions between mutated colicins and immunity proteins were monitored and interactions sites were identified. |
