Description 15 wild barley (H. v. ssp. spontaneum) sources of resistance to powdery mildew were studied in order to localize the resistance genes on barley genetic map using DNA markers and saturate the corresponding chromosome regions with more tightly linked markers. The analyses were performed in F2 populations. Microsatellite markers were used for mapping and for the regions with few polymorphic markers, 11 STS (sequence-tagged site) markers were converted into new CAPS (cleaved amplified polymorphic sequence) markers; their polymorphism was tested after cleavage of the PCR products with suitable restriction enzymes. The data from F2 plants of all genotypes were analysed in the MapManager QTX program to assign the order and distances between the markers and the probable positions of the resistance genes. In Tiffany x the accession PI466197, 38 out of 117 SSR and 3 out of 4 newly developed CAPS markers on chromosome 2H were used. One resistance gene was located between the markers Bmac0213 and MGB402 on chromosome 1H (in the Mla locus) and the other, newly identified gene was located on chromosome 2H between Bmac0134 and MWG878, 4 cM from Bmac0134.
