Description | Electrospray ionization (ESI) and matrix-assisted laser desorption/ionization (MALDI) are preferred ionization techniques in analysis of polymers. While ESI is usually chosen for on-line coupling of column separations to MS, MALDI is used for MS screening of large number of samples. Certain features of MALDI, however, make it more attractive in some applications: MALDI spectra, dominated by singly charged ions, are simpler and solid samples prepared on MALDI targets can be archived. This lecture will give an overview of approaches for coupling of microLC and CE with MALDI MS. The power of separation coupled a TOF/TOF instrument will be demonstrated on sensitive multidimensional analysis of complex biological samples. In conjunction with TOF mass spectrometer and high repetition rate lasers, extremely fast analyses can be achieved. The potential for high throughput MS analysis will be discussed on example of on-line and off-line analysis. |
