Description | In our laboratory, early stages of ischemia are studied using various models and methods. The aim of this study was to verify our preconditioning model of rabbit isolated heart perfused according to Langendorff loaded with voltage-sensitive dye. The hearts of seven New Zealand rabbits were perfused with Krebs-Henseleit solution, loaded with di-4-ANEPPS and washed out. Monophasic action potentials were recorded by optical probe, 3D-electrogram by touch-less method and mean coronary flow by collecting the perfusate. Three successive periods of global (flow) ischemia followed by reperfusion were introduced (10 min each). Typical changes of the electrogram shape were present (e.g. ST elevation), together with few arrhythmias. Longer periods of ischemia are needed in order to verify this model in myocardium affected by voltage-sensitive dye. |
