Description Colon carcinoma belongs to the most frequent tumor diseases in industrial countries. Development of the colon tumors is associated with mutations occurring in protooncogenes and tumor suppressor genes of colon cells. The c-myb protooncogene is one of the genes involved in colorectal cancerogenesis. It codes for the c-Myb transcription factor. The set of the Myb-targeted genes includes the cox-2 gene coding for cyclooxygenase 2. Cells of colon tumors often overexpress Cox-2. The aim of this study was to determine the effect of c-Myb overexpression and c-Myb silencing, respectively, on apoptosis of HCT116 and HT-29 colon carcinoma cells and to reveal whether Cox-2 acts as mediator of effects controled by c-Myb. We show, that c-Myb suppresses cis-platin/TRAIL-induced apoptosis of colon carcinoma cells in Cox-2 independent manner.
