Searching for Genes of Lactococcus lactis subsp lactis Encoding the Bacteriocin Nisin using DNA/DNA Hybridisation

Description In this work, PCR primers P8 and P9 were used for amplification of a 320 bp long PCR product specific to the nisin gene. The PCR product was labelled with digoxigenine during amplification and used as a DNA probe for the screening of homologous DNA sequences in 7 Lactococcus lactis subsp. lactis strains from the Culture Collection of Dairy Microorganisms (CCDM). Dot blot hybridisation and hybridisation of colonies were used for DNA/DNA hybridisation. It was shown that 6 tested strains of Lactococcus lactis subsp. lactis have genes encoding nisin. One strain had probably a defective gene encoding nisin.
