Description | A new method for determination of trace elements in submicroliter sample volumes using substrate-assisted laser desorption inductively-coupled plasma mass spectrometry is presented. Well-defined volumes of samples are deposited on absorbing substrate, desorbed with pulses of 213-nm laser at relatively low laser power density and analyzed with ICP MS. SALD ICP MS can be used for the determination of trace elements in minute amounts of biological samples as demonstrated on the determination of copper in myeloid leukemia cells to investigate the role of copper ions in cytotoxicity regulation with disulfiram . Furthermore, biological samples, such as metallothioneins can be analyzed by matrix-assisted laser desorption/ionization mass spectrometry (MALDI MS) after addition of matrix to the sample. Complementary SALD ICP MS and MALDI MS techniques provide information on both metal content and protein mass or identity from the same sample. |
