Description | The energetically important metabolites play a key role in metabolism. Their monitoring allows verifying or predicting the physiological state of the cell. On this account, the main aim of this study was to optimize capillary electrophoretic method for selective and rapid determination of purine and pyrimidine nucleotides, adenine coenzymes and Acetyl CoA from bacterium Paracoccus denitrificans. In order to improve the concentration sensitivity, the capillary zone electrophoresis (CZE) was combined with the on-line preconcentration technique – field enhanced sample stacking. First of all the method was optimized through the use of metabolite standards and then this method was used for analysing of cell extract. The separation conditions were necessary to adjust in order to identify (or quantify in the case of metabolites over LOQ) all demanded and detectable metabolites. |
