Popis INTRODUCTION: The tooth development is a complex process influenced by reciprocal and precisely synchronized interactions between the oral epithelium and mesenchyme. It is initiated and controlled by large amount of signal molecules, the most explored are BMP, WNT, FGF and HH pathways. Here, we selected gene Satb2 for our analysis, which is involved in transcriptional regulation and chromatin remodeling. Expression of this gene during early development is restricted to the medial portion of the first to fourth pharyngeal arches. It also plays role in craniofacial patterning and development, palate formation, and osteoblast differentiation and maturation. Here, we focused on the analysis of SATB2 expression during tooth development. METHODS: Protein expression of SATB2 was analyzed by immunohistochemical detection at prenatal (E11.5-E18) as well as postnatal (P0-P10) stages during odontogenesis. For QPCR analysis, tissues were dissected from the rostral and caudal areas of mouse mandibles at different stages of prenatal development to analyze regional differences in time at mRNA level. RESULTS: SATB2 demonstrated distinct region-specific expression in the early lower jaw and in the orofacial area. Moreover, SATB2 was expressed in the ameloblasts and odontoblasts during prenatal and postnatal stages. Gene expression analysis by QPCR revealed significant differences in Satb2 expression between incisor and molar areas of the mandible with higher expression in the rostral zone during early embryonic stages. CONCLUSIONS: Spatiotemporal dynamics in SATB2 expression in orofacial region and during odontogenesis suggest a distinct role of this protein during prenatal development.
