Popis The v-myb oncogene is efficiently expressed in BM2 monoblasts thus blocking their terminal differentiation steps. Histone acetylation plays an important role in regulation of transcription and is particularly relevant to regulation and pathology of hematopoiesis. Recently, we have shown that inhibitor of histone deacetylases trichostatin A (TSA) causes histone hyperacetylation and differentiation of BM2 cells into macrophage polycaryons. In this work we used another approach to stimulate histone acetylation in BM2 cells. We prepared two variants of BM2 cells ectopically expressing two histone acetyl transferases CBP and p300. The expression of CBP as well as p300 itself did not affect the phenotype of BM2 cells, but it enhanced their response to the differentiation inducers TPA and TSA. Although CBP and p300 are closely related proteins, many differences in their function have been described. We noted differences in kinetics of differentiation induced by CBP and p300 in BM2 cells.
