Popis | Staphylococcus aureus is a major human and veterinary pathogen causing local and systemic infections. The pathogenicity of S. aureus depends on production of virulent factors and contents of resistant determinants to antimicrobial agents. These virulent and resistant factors are carried on mobile genetic elements (MGE). Different types of MGE comprise 15 % of genome S. aureus and can be horizontal transferred. The transduction mediated by temperate staphylococcal bacteriophages of serological group B is one of the horizontal gene transfer mechanisms.The aim of the project is detection and quantification of bacterial genes on MGE, which are transferred by transducing phage particles. The work is focused on staphylococcal chromosomal cassettes (SCCmec), pathogenicity islands (SaPIs), plasmids and chromosomal genes encoding virulent factors. qPCR was used for the quantification and detection of genes. It is more sensitive and has higher resolution level than end-point PCR. The plasmid copy number in S. aureus, resistant genes on plasmids, the parts of SCCmec and SaPIs in transducing phage particles were determined by qPCR. The results of this project contribute to explanation of mechanism and frequency of horizontal gene transfer by transducing phage particles. |
