Popis | Gold nanoparticles (GNPs) are the object of interest in the field of nanotechnology due to their unique electronic, structural, optical, magnetic, thermal, and catalytic properties. They are extensively applied in the construction of electrochemical sensors and biosensors. The aim of this contribution is to characterize GNPs of both sizes (diameters of 5nm and 10 nm) in buffered and non-buffered aqueous solutions with the change of ionic strength by spectroscopic methods (UV/ Vis, photon correlation spectroscopy), and to investigate the GNP–aptamer–thrombin system as an application of GNPs in a sensitive aptasensor. For the immobilization the thrombin-binding aptamer (TBA) with the sequence GGT TGG TGT GGT TGG on the GNP surface was modified by a sulfhydryl group. The study showed that the spectral characterization of GNPs depends strongly not only on the aptamer–thrombin conjugate but also on the GNP size and solution composition (buffer, ions and their concentrations). |
