The DNA mismatch repair (MMR) system corrects mispaired or unpaired bases as well as other DNA lesions, thus plays a critical role in sensitizing both prokaryotic and eukaryotic cells to the clinically potent anticancer drug cisplatin. It is thought to mediate cytotoxicity through recognition of cisplatin DNA lesions. In this study, interaction of MutS protein with site-specifically modified DNA oligonucleotides containing cisplatin cross-links will be investigated using mainly NMR spectrosopy. With the use of the latest advancements in the field and utilizing molecular modeling, complex formed by the DNA adduct with the MutS will be determined. The scope of this structural study is helping to understand the prerequisite step - recognition of the DNA cisplatin lesions - in the chain of events leading to cisplatin cytotoxicity mediated by MMR.
