Description Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI TOF MS) allows rapid determination of the oligosaccharides masses, oligomer dispersion and also has a potential for the quantitative analysis. In this study, method for MS quantification of food oligosaccharides using a standard addition of a pure monodisperse tetraoligosaccharide nystose with minimal optimisation including sampling and laser energy, was developed. Relative quantification was based on evaluation of change in peak intensity of four raftilose oligomers with degree of polymeration from 3 to 6 and assumption of constant oligomer dispersion profile. Raftilose content was evaluated with two mathematical approaches, conventional calculation and using artificial neural networks (ANN).
