Determination of deoxynivalenol levels in barley, malt and intermediate products of malting process by high performance liquid chromatography and MALDI-TOF mass spectrometry

Description The deoxynivalenol (DON) is the most frequently detected mycotoxin in grain samples and their subsequent food products (beer); associated with primary gushing. The extracted samples were analysed using RP-HPLC - using mobile phase AcN/H2O (9:1); flow rate 0.6 ml.min-1; column temperature at 25 C; sample dosage 20 ul; detected at 220 nm. The LOD and the LOQ for DON was 0.16 and 0.53 ug.ml-1, respectively 127.8 and 426.2 ug.kg-1. Suitable matrix for MALDI-TOF MS analysis were found matrix of sodium azide and diamond suspension, quick and dirty spotting of sample and matrix solution 3:1. Positive reflector mode and PE with optimum mass set to 300 Da were used. Adducts of DON were observed. The LOD for DON was 0.6 ug.ml-1, resp. 480.0 ug.kg-1. In this study we found out that the levels of DON during malting process increased (maybe caused by DON releasing from enzymatic conjugates). The correlation between DON content and the level of gushing was negligible.
